There are many Nemesis bosses spawns in Tibia, which spawn randomly within an interval of several days. These bosses spawn acts like a raid. For example: Sir Valorcrest, Yaga the Crone, Zarabustor.

Certain monsters in Tibia will not spawn until the spawn area is clear of player characters (For example, a rat)

Will a character remaining online around the boss's spawn, will block the boss from spawning?

The Nemesis bosses which spawn randomly every few days are often sought after by boss hunters, either for completing the Bosstiary or acquiring rare loot which they drop.

For this reason you will often see these spawns with players coming in, or lower level characters logging in and out at the spawn location.

This often raises a question, whether these players may be unintentionally delaying or blocking the boss's spawn by their presence.

From my own experience, I encountered these bosses inside their spawn during the very moment they spawned. This means you cannot block their spawn. They act similar to a raid (When a message appears informing of monsters spawning next to a city, for example). A raid cannot be blocked, and the monsters instead spawn around the players at the location.
