Let's say, I have 3500 Tibia Coins in total (including 250 transferable Tibia Coins) and I win a character bid of 4000 Tibia Coins. Will the non-transferable Coins cover the payment and I'll have to get another 500 Tibia Coins to buy the character, or just the transferable 250 TC  will be taken into account and I'll have to get another 3750 transferable Tibia Coins? Thanks in advance.

You'll need to purchase more transferable coins. Please see my answer below.

When buying a character it has to be made with transferable coins only.

"Security plays a crucial role in character auctions. Hence, using an authenticator is mandatory to partake in character auctions and payments have to be made using transferable Tibia Coins only." Source: Character Auctions

When you successfully sell a character you might be receiving partly or completely non-transferable tibia coins.

"Note that the Tibia Coins you receive for selling a character may be partly or completely non-transferable initially as a matter of fraud protection. Non-transferable coins gained through a character sale become transferable at the latest after 120 days." Source: Character Auction Overview and FAQ
